For some reason, when you go to save the brother and you knock on the door to convince him to come out, it says for you to clear the room full of infected. Use the drawbridge to get to the building through the upper door near the roof (picture above). After entering the building, turn left and reach the metal gate. You can go into the room and start exploring it.
